【摘要】:Increasedemphasisisbeingplacedonprotectingwetlands(6)andseveralagenciesincludingU.S.EnvironmentalProtectionAgencyandEnvironmentCanadaarebeginningtorequire,fortheregistrationofpesticides,dataregardingtoxicityoftestmaterialstorootedaquaticvascularplants(7,8,9).Muchresearchisbeingconductedwithvascularplants,bothterrestrialandaquatic(10),however,protocolsforphytotoxicitytestingwithfreshwateremergentmacrophytesstillarenotwelldefined.Thisguideisdesignedtoassesspotentialdetrimentaleffectsofwatersolublechemicalsubstancesonselectedsurrogatespeciesoffreshwateremergentmacrophytes.Thisguidefocusesondiminishmentofchlorophyllcontentinleavesasthemeasurableendpoint,however,notallchemicalsaffectchlorophyllproduction.DryweightcanbeusedastheendpointforO.sativa,however,exposuretimesmayneedtobeextendedtodetecttreatmentdifferences.Dryweightisnotarecommendedendpointforanyofthetestspeciesstartedasrhizomesortubers.Otherendpoints,suchasperoxidaseactivity(11)orchlorophyllfluorescence(12)couldpossiblybeused.Thisguidecouldbeusedtoprovideearlyindicationofpotentialproblems,identifyhazardoussubstancesbeforecontaminationofwetlandsoccurs,andestablishx201C;marginsofsafetyx201D;forspecificchemicalswithinwetlands(seeGuideE1023).Thisguideisnotdesignedtoreplacefieldassessmentsorotheraquatictestingprocedures.Itisdesignedtocomplimentsuchtesting,sothatamorecompleteassessmentispossible.1.1Thistestguideisdesignedtogivegeneralguidanceforassessingthepotentialphytotoxicityofwatersolubletestmaterialtofreshwateremergentmacrophytes.1.2Thisrenewaltestcontinuouslyexposesselectedplantspecies,growinginsediment,tovariousconcentrationsoftestmaterial,dissolvedinanutrientsolution.1.3ThistestguideisbasedontheToxicSubstancesControlAct(TSCA)guidelinesforconductingtoxicitytestswithterrestrialplants(1)andisapplicabletomostwatersolublechemicals,eitherindividuallyorinformulations,commercialproducts,orknownmixtures(seeGuidesE1193andE1598).Withslightmodificationstheprocedurealsomightbeusedforeffluents(seeGuideE1192).1.4ResultsfromthistoxicitytestcanbeusedtoreportanIC50orNOEC(seeSection3)basedontheconcentrationofchlorophyllextractedfromtheplants(seeGuidesD3731andE1218).Insomesituations,itmightbenecessarytoonlytestatoneconcentrationtodeterminewhetherornotthatspecificconcentrationistoxictotheplants.1.5Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.SpecifichazardstatementsaregiveninSection7.
